幽门螺杆菌(Helicobacter pylori)鞭毛蛋白A(FlaA)可作为临床诊断标志物,但目前尚无识别FlaA蛋白的特异性抗体.该研究构建了HpflaA基因表达质粒,表达并纯化了HpFlaA重组蛋白,并以此重组蛋白为抗原免疫BALB/c小鼠,通过杂交瘤细胞技术制备HpFlaA单克隆抗体,最后对抗体的亚型、效价、纯度、亲和力和特异性等进行鉴定.共获得5株能稳定分泌单克隆抗体的杂交瘤细胞株,分别命名为2G2、2G10、3E2、3E4和4H3.这5株细胞分型均为IgG1型且抗体效价均超过1:105,亲和常数均达到107以上.经间接酶联免疫吸附实验和蛋白质免疫印记实验验证,单克隆抗体能特异性识别HpFlaA.成功制备出高亲和力、高效价、特异性好的HpFlaA单克隆抗体,具有潜在临床应用价值.
Flagellin A(FlaA)serves as a diagnostic marker for Helicobacter pylori,but the specific antibody against FlaA is not available so far in our country.In this study,we generated the expression plasmid for HpFlaA gene,expressed and purified the recombinant HpFlaA protein.This purified HpFlaA was used to immunize BALB/c mice to generate the monoclonal antibody through hybridoma technology.Finally,we characterized the subtype of immunoglobin,and determined the purity,the titer,the affinity and the specificity of the antibody by indirect ELISA experiments.In this study,5 hybridoma cell lines capable of stably secreting the monoclonal antibody were obtained and named as 2G2,2G10,3E2,3E4 and 4H3.The antibody subtypes of all 5 strains were determined as IgG1,the titers were over 1:105 for all 5 monoclonal antibodies,and the binding affinity to antigen was over 107.Indirect ELISA and Western blot confirmed that the monoclonal antibodies can specifically recognize HpFlaA.We successfully generated the monoclonal antibody specifically against HpFlaA with high-affinity,high-titer and high-specificity,which has potential significance for clinical diagnosis.
