采用乌氏黏度计测定不同pH条件下牛血清白蛋白(BSA)的黏度,运用Wolf方程描述BSA的黏度变化行为.详细讨论了不同pH条件下BSA的特性黏数(intrinsic viscosity,[η])与其构象的关系,并分析了BSA比特性黏数(specific intrinsic viscosity,{η})的组成变化.研究结果表明,采用这种新型的黏度法研究球状蛋白质的构象或溶液性质是科学、可行的.

The viscosities of bovine serum albuminin(BSA)in different pH buffers were measured using Ubbelohde capillary viscometer,and the data were evaluated by Wolf equation to describe the composition dependences of BSA.The intrinsic viscosities([η])of BSA under different pH were correlated with its conformation,and the composition dependences of specific intrinsic viscosities({η})of BSA were carefully analyzed.Our results indicate that the new viscometric method can be applied in the study of conformation and solution properties of spherical protein.
